These cells represent potential biomarkers with which to follow disease progression, as well as providing important biological information into the process of metastasis. Examples will be provided, including the monitoring of tumor genotypes in EGFR-mutant lung cancer, as well as the molecular characterization of CTCs from patients with localized and metastastic prostate cancer. His laboratory interests have focused on the area of cancer genetics, including the etiology of the pediatric kidney cancer Wilms tumor and genetic predisposition to breast cancer. Recently, his laboratory reported that lung cancers with activating mutations in the epidermal growth factor receptor (EGFR) are uniquely sensitive to tyrosine kinase inhibitors that target this receptor. as opposed to chemotherapy, surgery, and standard radiation? I know that there has to be a safer cancer treatment out there unlike chemotherapy or surgery and standard radiation that is covered by my health insurance or an alternative treatment for cancer. INTRODUCTION
Mesenchymal-epithelial transition (Met) tyrosine kinase is the receptor for hepatocyte growth factor (HGF), which is normally active in wound healing, liver regeneration, and embryonic development. However, the Met pathway is one of the most frequently dysregulated pathways in human cancer. Met dysregulation leads to constitutive activation of its tyrosine kinase activity thus increasing the downstream signals that lead to cell survival, proliferation, and migration. Patients with upregulated Met tend to have aggressive, chemo-resistant cancers and poor prognoses. For these reasons, the pharmaceutical industry has expended much time and effort to identify potential Met tyrosine kinase inhibitors as potential therapeutics for these cancers.
POTENTIAL THERAPEUTICS
CrizotinibThe drug Crizotinib has been discussed previously in its role as an EML4-ALK inhibitor for treating non-small cell lung carcinoma. However, Crizotinib is also a Met-inhibitor and shows promise in several other forms of histological cancer associated with Met-dysregulation.
ForetinibForetinib is an inhibitor of both Met and vascular endothelial growth factor receptor (VEGFR). Foretinib completed Phase I clinical trials in 2010 and showed potential in treating aggressive solid tumors. Phase II clinical trials are currently underway testing its efficacy in both lung and breast cancer.
CabozantinibLike foretinib, Cabozantinib inhibits both Met and VEGFR. It was granted orphan drug status by the FDA in 2011. The orphan drug status is available to encourage pharmaceutical companies to develop drugs for diseases, known as orphan diseases, which have a small market because they are relatively rare. Orphan drugs proceed through their development along the same regulatory pathways as other drugs with the exception that some statistical burdens may be lessened in recognition of the limited patient pool. Furthermore, other financial incentives are also linked to orphan drugs such as tax incentives and extended patent protection. These exceptions are in place to encourage the pharmaceutical industry to develop drugs for these rare diseases, which, by definition, would have a small market share and be relatively unprofitable.
Cabozanitib is currently in clinical trials for treatment of prostate, ovarian, brain, melanoma, breast, non-small cell lung, hepatocellular, kidney, and medullary thyroid cancer.
SUMMARY
The Met tyrosine kinase is one of the most dysregulated pathways in many cancers. The use of c-Met inhibitors with other therapeutic agents could be crucial for overcoming potential resistance as well as for improving overall clinical benefit in multiple types of cancer. Therefore, for more than two decades, investigators have been exploring Met inhibitors as potential cancer treatments. Three drugs currently in development are crizotinib, foretinib, and cabozantinib, all of which are currently in early phase clinical trials for treatment of several different cancers. Met inhibitors may prove to be a targeted type therapy with reduced toxicity, that are effective at halting metastasis and improving prognosis in many types of previously resistant tumors.

In the last several years, as researchers expand their search for novel anticancer plants and compounds, certain essential oils have been repeatedly turning up in published reports as being potential future therapies for cancer treatment. Essential oils are particular kinds of extracts from plants — they are what give plants their aromas. More alcohol-like than oil-like, these compounds easily evaporate and are readily detected by our olfactory systems. Less known is that these volatile compounds are often highly complex, and can be very unique from plant to plant. A great many of these oils have been the subject of medical research in several fields, and are noted for their anti-viral, anti-bacterial, and inflammation-reducing actions. Now, their anticancer activities are coming to light as well.
Two Outstanding Essential Oils Researched for Anticancer Activity
The essential oils distilled from Frankincense resin and from the leaves of Lemongrass have been the subject of more scientific inquiry regarding their efficacy as cancer treatments than any other oils. Why this is so is somewhat uncertain, though both these plants have a long history of use as medicines. The term Frankincense refers specifically to the resin or sap of one of several species of Olibanum tree, native to east Africa and India. The resin even has biblical significance, being one of the gifts of the Magi to the newly-born Jesus — it was at that time a very costly medicine, incense and perfume. Lemongrass has been used in more tropical environs for its anti-infectious properties, with detoxifying effects several bodily systems. It is said to aid in digestion, circulation, and in clearing the skin.
Modern Research Proves Ancient Medicine’s Potential
Frankincense essential oil is noted in “The Complete Guide to Aromatherapy” by Salvatore Battaglia as an immuno-stimulant, an oil that does not eradicate viruses or bacteria directly, but improves the body’s own natural immunity. A look at the research performed over the last few years is quite impressive: several pages of abstracts are returned when searching specifically for research regarding its potential as a cancer therapy. Many of these investigations have found the same result: Frankincense (and actually other essential oils that have similar effects) apparently targets only cancerous cells, leaving otherwise healthy cells unaffected.
Research is focusing on the chemical components of the essential oil unique to frankincense: the Boswellic acids. These natural compounds have been thoroughly studied for their anti-inflammatory activity, with a significant body of positive results. Boswellic acids can be used to reduce pain and inflammation in arthritic joints, and has even been shown to improve the texture and appearance of skin that may be prematurely aged due to sun overexposure. It is these same Boswellic acids that have been shown to induce natural cell death in cancerous cells — it is one of the features of cancer that natural cell death does not occur, and the cancerous cells then in essence grow unabated throughout the body.
Impressive Targeting of Cancer Cells
The conclusion of a study published in the journal of Complementary and Alternative Medicine, March 2009, by researchers at the University of Oklahoma was “Frankincense oil appears to distinguish cancerous from normal bladder cells, and suppress cancer cell viability”. This is truly a profound result, when considering what modern chemotherapy does — reduces the viability of all cells in and surrounding a tumor, with the hope that healthy cells live through the process in significant enough numbers that the patient recovers. Similar results have been published for a great many kinds of cancer: melanoma, leukemia, liver, colon and prostate, and anecdotal reports have supported its use in the treatment of breast cancer as well.
Lemongrass: Tropical Healer
The first result of a search for ‘lemongrass’ and ‘cancer’ is a paper titled “Anticancer activity of an essential oil from Cymbopogon flexuosus” (Cf is a popular species of Lemongrass, though other research also shows similar effects from Cymbopogon citratus). This study published in the May 2009 ‘Chemico-biological Interactions’, performed at the Indian Institute of Integrative Medicine, examined both in-vitro (essentially “in the test tube”)and in-vivo (in the body) anticancer activity of Lemongrass essential oil. The oil was actually evaluated for efficacy against 12 cancer cell lines in-vitro, and 2 in-vivo. The results were no less than astounding. Lemongrass was effective at inhibiting proliferation of all the cell lines, though with significant variation in the concentrations required for this effect. The lowest concentrations of Lemongrass were needed to inhibit growth of colon cancer and neuroblastoma cells in-vitro (a type of cancer of the nerve cells occurring in children and infants). Using mouse models, lemongrass essential oil was effective at limiting proliferation of a type of breast cancer and a connective-tissue cancer.
Israeli researchers, publishing their results in the May 2007 Planta Medica, discovered similar results focusing on a primary constituent of Lemongrass essential oil: Citral. This natural constituent adds the ‘lemony’ aroma to many essential oils, including of course Lemon, Melissa, and Lemon Verbena. Citral was noted to also show cancer cell specificity (liver cancer cells were evaluated in this case), causing only the cancer cells to die off. It appears that lemongrass essential oil, like Frankincense, also induces natural cell death in cancer cells, sparing normal healthy cells in the process.
These studies (and others available through medical research databases) are indeed promising — most noting in some way that the low toxicity, availability and cancer-cell specificity of these natural compounds warrant further investigation. Because of the current climate regarding funding of alternative therapies, it will take either a government wishing to significantly save on health care costs, or a large grass roots effort, to further the research to a point where specific therapeutic protocols are developed. In the meantime, there are many respected, well-educated health professionals that may be willing to offer advice for those wishing to learn more on how Frankincense and/or Lemongrass might be employed as preventative measures.
